What Is PT-141?

PT-141 (bremelanotide) is a synthetic peptide derived from the melanocortin system. Unlike PDE5 inhibitors that work through vascular mechanisms, PT-141 acts directly on the central nervous system by activating melanocortin receptors (MC3R and MC4R) in the hypothalamus.

The FDA approved bremelanotide (branded as Vyleesi) in 2019 for the treatment of hypoactive sexual desire disorder (HSDD) in premenopausal women. However, research and anecdotal reports suggest it may be effective for both men and women. It is administered as needed via subcutaneous injection, typically 45-60 minutes before anticipated activity.

How to Store PT-141

Before reconstitution: Store lyophilized (powder) pt-141 in a cool, dry place away from direct sunlight. Refrigeration is ideal for long-term storage of unreconstituted peptides.

After reconstitution: Store the reconstituted vial upright in a refrigerator at 36-46°F (2-8°C). Do not freeze. The solution typically remains stable for 4-6 weeks. Always swab the vial stopper with an alcohol pad before each use.

How far in advance should I take PT-141?+
Most people inject PT-141 approximately 45-60 minutes before anticipated activity. Effects can last 6-12 hours depending on individual response. Some users report effects beginning as early as 15-30 minutes post-injection.
Can men use PT-141?+
While the FDA-approved version (Vyleesi) is indicated for premenopausal women, PT-141 has been studied in both men and women. Research in men with erectile dysfunction showed promising results, particularly in cases where PDE5 inhibitors were ineffective, as PT-141 works through a completely different (central nervous system) mechanism.
Can PT-141 be used with PDE5 inhibitors?+
Some individuals combine PT-141 with PDE5 inhibitors, but this should only be done under medical supervision. Both can affect blood pressure, and the combination has not been extensively studied for safety. Start with the lowest effective dose of each if combining.
How many times per month can I use PT-141?+
The FDA recommends no more than 8 doses per month, with at least 24 hours between doses. Using PT-141 too frequently may lead to tolerance (reduced effectiveness) and increases the risk of side effects such as skin darkening.
Why does PT-141 cause nausea?+
Nausea is related to PT-141's activation of melanocortin receptors in the brain, which also influence the nausea/vomiting center. It is usually mild and self-limiting. Taking an anti-nausea medication 30 minutes before PT-141 or starting with a lower dose can help. Nausea tends to decrease with subsequent uses.
